    In recent years there have been an extraordinary number of proposals for comprehensive and intensive changes in school structure, finance, and administration, one indication that this is a time of unprecedent change for schools. One of the forces driving this intense focus on school change is the growing national concern about youth issues and problems. This paper notes the myriad of add-on programs that compete for space in the school day and takes the position that, rather than being seen as merely additive,education and services should be viewed as interactive, and essential to the formation of new effective strategies for educational success.The paper contends that if the additive view remains the sole model of school-community collaboration, it will be detrimental to school reform. Rather, there must be collaboration to broaden the knowledge base and schools must seek partners to build a broader vision of what education should be and can accomplish. The paper examines what needs to change, arguing that what is needed is a shift to thinking that supporting youth development is the most effective strategy for the prevention of youth problems and the achievement of educational goals. It proposes a definition of youth development, considers expanding definitions of positive youth outcomes, and focuses on two components of youth development--competencies and needs--and their interrelationship. The document concludes that the broader goals and strategies of youth development must become central to the educational mission

    Expect the Unexpected When Teaching Probability

    Probability has recently made its way into many textbook series and standards documents (NCTM, 2000; NGA, 2010). When students engage in probability problem solving many unexpected situations can arise due to the counterintuitive nature of probability concepts. These situations can be difficult for students and challenging for teachers to analyse during teaching. Recently, as facilitators of a Mathematics Science Partnership grant workshop on probability, we had the opportunity to engage middle school teachers in professional development workshops as well as in their classrooms. In this article, we discuss a rich probability task used with these teachers along with two scenarios that represent challenging aspects of probability for students, and challenging teaching. In these two scenarios, we explain the underlying probabilistic concepts that proved difficult for students. For each probability challenge, we discuss how the process of analysing student thinking can inform teaching strategies that may guide student conceptual development

    Moving Beyond Where and When to the People and Practices That Impact Youth

    Descriptors like after-school and out-of-school make it easy for funders and families to understand the basics of what the youth development field offers—places for young people to go when they are not in school. These descriptors, however, do little to promote the youth work profession. Youth work is best practiced in settings that prioritize relationships, promote social and community engagement, and provide opportunities for interest-based skill building and exploration. The value youth workers bring to their practice, as the archive of articles in this journal demonstrates, is not in the basic descriptors of the setting, but in the design of the learning environments. This article explores the need for youth work professionals to use, contribute to, and share our knowledge base within and outside of the field

    Pharmacokinetics, safety and tolerability of olaparib and temozolomide for recurrent glioblastoma: results of the phase I OPARATIC trial

    Background: The poly(ADP-ribose) polymerase (PARP) inhibitor olaparib potentiated radiation and temozolomide chemotherapy in pre-clinical glioblastoma models but brain penetration was poor. Clinically, PARP inhibitors exacerbate the hematological side-effects of temozolomide. The OPARATIC trial was conducted to measure penetration of recurrent glioblastoma by olaparib, and assess the safety and tolerability of its combination with temozolomide. Methods: Pre-clinical pharmacokinetic studies evaluated olaparib tissue distribution in rats and tumor-bearing mice. Adult patients with recurrent glioblastoma received various doses and schedules of olaparib and low-dose temozolomide in a 3+3 design. Suitable patients received olaparib prior to neurosurgical resection; olaparib concentrations in plasma, tumour core and tumour margin specimens were measured by mass spectrometry. A dose expansion cohort tested tolerability and efficacy of the recommended phase II dose (RP2D). Radiosensitizing effects of olaparib were measured by clonogenic survival in glioblastoma cell lines. Results: Olaparib was a substrate for multi-drug resistance protein-1 and showed no brain penetration in rats but was detected in orthotopic glioblastoma xenografts. Clinically, olaparib was detected in 71/71 tumor core specimens (27 patients, median 496nM) and 21/21 tumor margin specimens (9 patients, median 512.3nM). Olaparib exacerbated TMZ-related hematological toxicity, necessitating intermittent dosing. RP2D was olaparib 150mg (3 days/week) with TMZ 75mg/m2 daily for 42 days. Fourteen (36%) of 39 evaluable patients were progression-free at 6 months. Olaparib radiosensitized six glioblastoma cell lines at clinically relevant concentrations of 100 and 500 nM. Conclusions: Olaparib reliably penetrates recurrent glioblastoma at radiosensitizing concentrations, supporting further clinical development and highlighting the need for better pre-clinical models

    Integrating team science into interdisciplinary graduate education: an exploration of the SESYNC Graduate Pursuit

    Complex socio-environmental challenges require interdisciplinary, team-based research capacity. Graduate students are fundamental to building such capacity, yet formal opportunities for graduate students to develop these capacities and skills are uncommon. This paper presents an assessment of the Graduate Pursuit (GP) program, a formal interdisciplinary team science graduate research and training program administered by the National Socio-Environmental Synthesis Center (SESYNC). Quantitative and qualitative assessment of the program’s first cohort revealed that participants became significantly more comfortable with interdisciplinary research and team science approaches, increased their capacity to work across disciplines, and were enabled to produce tangible research outcomes. Qualitative analysis of four themes—(1) discipline, specialization, and shared purpose, (2) interpersonal skills and personality, (3) communication and teamwork, and (4) perceived costs and benefits—encompass participants’ positive and negative experiences and support findings from past assessments. The findings also identify challenges and benefits related to individual personality traits and team personality orientation, the importance of perceiving a sense of autonomy and independence, and the benefit of graduate training programs independent of the university and graduate program environment

    A comprehensive analysis of rare genetic variation in amyotrophic lateral sclerosis in the UK.

    Amyotrophic lateral sclerosis is a progressive neurodegenerative disease of motor neurons. About 25 genes have been verified as relevant to the disease process, with rare and common variation implicated. We used next generation sequencing and repeat sizing to comprehensively assay genetic variation in a panel of known amyotrophic lateral sclerosis genes in 1126 patient samples and 613 controls. About 10% of patients were predicted to carry a pathological expansion of the C9orf72 gene. We found an increased burden of rare variants in patients within the untranslated regions of known disease-causing genes, driven by SOD1, TARDBP, FUS, VCP, OPTN and UBQLN2. We found 11 patients (1%) carried more than one pathogenic variant (P = 0.001) consistent with an oligogenic basis of amyotrophic lateral sclerosis. These findings show that the genetic architecture of amyotrophic lateral sclerosis is complex and that variation in the regulatory regions of associated genes may be important in disease pathogenesis
